BTC2 exhibited an average IC50 value of 0.54μM, a desirable selectivity index of >2, inhibited the migration of ER+ and TN breast cancer cells, and displayed anti-cancer stem cell activity. We demonstrate that BTC2 induced DNA double strand breaks (increased levels of γ-H2AX) and activated the p-ATM/p-CHK2 and p-p38/MAPK pathways resulting in S- and G2/M-phase cell cycle arrests. Importantly, BTC2 sensitized breast cancer cells by triggering the intrinsic (cleaved caspase 9) and extrinsic (cleaved caspase 8) apoptotic as well as necroptotic (p-RIP3 and p-MLKL) cell death pathways and inhibiting autophagy and its pro-survival role. Furthermore, in the xenograft in vivo model, BTC2 displayed limited toxicity and arrested the tumor growth of breast cancer cells over a 9-day period in a manner comparable to that of the pos. control drug, paclitaxel. BTC2 thus displayed promising anti-breast cancer activity.

A crystal structure of [Pd(PyDT)(ppy)] reveals that the PyDT ligand is bonded as a bidentate chelate. Reaction of trans-[PdCl2(DMSO)2] with sodium benzisothiazolinate (NaBit), followed by NH4(PyDT) afford Na[Pd(PyDT)(N-Bit)2]. Diphosphine adducts [Pd(PyDT)2{Ph2P(CH2)nPPh2}] {n = 2-4 or (CH2)n = (C5H4)2Fe}, can be prepared in good yield upon addition of the diphosphine to [Pd(PyDT)2]. Reaction of trans-[PdCl2(PPh3)2] with sodium saccharinate (Nasac) followed by NH4(PyDT) afford trans-[Pd(PyDT)(N-sac)(PPh3)2]. The prepared complexes were characterized by elemental anal., IR, 1H, 31P NMR spectroscopic data and conductivity measurements.

In search of a connection between prebiotic peptide chemistry and lipid compartments, the reaction of a 5(4H)-oxazolone with leucinamide was extensively explored under buffered aqueous conditions, where diverse amphiphiles and surfactants could form supramolecular assemblies. Significant increases in yield and changes in stereoselectivity were observed when fatty acids exceeded their critical aggregation concentration, self-assembling into vesicles in particular. This effect does not take place below the fatty acid solubility limit, or when other anionic amphiphiles/surfactants are used. Data from fluorimetric and Langmuir trough assays, complementary to the main HPLC results reported here, demonstrate that the dipeptide product co-localizes with fatty acid bilayers and monolayers. Additional experiments in organic solvents suggest that acid-base catalysis operates at the water-aggregate interface, linked to the continuous proton exchange dynamics that fatty acids undergo at pH values around their effective pKa. These simple amphiphiles could therefore play a dual role as enhancers of peptide chemistry under prebiotic conditions, providing soft and hydrophobic organic domains through self-assembly and actively inducing catalysis at their interface with the aqueous environment. Our results support a systems chemistry approach to life’s origin.

We previously reported the discovery of several spirodiketopiperazine derivatives as potent CCR5 antagonists with anti-HIV activity. Herein, we describe in detail the identification of these lead compounds using a combinatorial chemistry approach.
