Fatty acids’ double role in the prebiotic formation of a hydrophobic dipeptide

In search of a connection between prebiotic peptide chemistry and lipid compartments, the reaction of a 5(4H)-oxazolone with leucinamide was extensively explored under buffered aqueous conditions, where diverse amphiphiles and surfactants could form supramolecular assemblies. Significant increases in yield and changes in stereoselectivity were observed when fatty acids exceeded their critical aggregation concentration, self-assembling into vesicles in particular. This effect does not take place below the fatty acid solubility limit, or when other anionic amphiphiles/surfactants are used. Data from fluorimetric and Langmuir trough assays, complementary to the main HPLC results reported here, demonstrate that the dipeptide product co-localizes with fatty acid bilayers and monolayers. Additional experiments in organic solvents suggest that acid-base catalysis operates at the water-aggregate interface, linked to the continuous proton exchange dynamics that fatty acids undergo at pH values around their effective pKa. These simple amphiphiles could therefore play a dual role as enhancers of peptide chemistry under prebiotic conditions, providing soft and hydrophobic organic domains through self-assembly and actively inducing catalysis at their interface with the aqueous environment. Our results support a systems chemistry approach to life’s origin.

Tribological properties of cross-linked oleophilic polymer brushes on diamond-like carbon films

Surface-initiated atom transfer radical copolymerization of hexyl methacrylate (HMA) and 3-ethyl-3-oxetanylmethyl methacrylate (OxMA) was carried out on the surfaces of block- and ring-type steel pieces covered with silicon-incorporated diamond-like carbon (DLC-Si) in order to generate an oleophilic copolymer brush layer at the outermost surface. The sample was then immersed in a 1% BF3OEt2 solution to form cross-linkages between oxetane groups in the polymer brush chains. The thickness of the polymer brush layer was confirmed to be 50 nm through transmission electron microscope images of the focused ion beam (FIB)-fabricated cross section. The friction properties of the composite films were evaluated using block-on-ring tests under a load of 49 N (130 MPa), using a base oil at 353 K for 30 min. Although the brush layer was partially scratched from the substrate surface during the friction test, the polymer brush-immobilized DLC-Si exhibited a low friction coefficient of 0.02, while the friction coefficient of the non-modified steel substrate was 0.12. It is supposed that the oleophilic polymer brush was swollen in the oil to form a stable lubrication layer, thus preventing the direct contact of the DLC-Si substrate. The dependency of the tribological properties on normal load, sliding velocity, wear depth, and the silicon content of the DLC-Si substrate was also investigated.

Synthesis and evaluation of [18F]fluororasagiline, a novel positron emission tomography (PET) radioligand for monoamine oxidase B (MAO-B)

The aim of this study was to synthesize and evaluate a novel fluorine-18 labeled analogue of rasagiline (6) as a PET radioligand for monoamine oxidase B (MAO-B). The corresponding non-radioactive fluorine-19 ligand, (1S,2S)-2-fluoro-N-(prop-2-yn-1-yl)indan-1-amine (4), was characterized in in vitro assays. The precursor compound (3aS,8aR)-3-(prop-2-yn-1-yl)-3,3a,8,8a- tetrahydroindeno[1,2-d][1,2,3]oxathiazole 2,2-dioxide (3) and reference standard 4 were synthesized in multi-step syntheses. Recombinant human MAO-B and MAO-A enzyme preparations were used in order to determine IC50 values for compound 4 by use of an enzymatic assay employing kynuramine as substrate. Radiolabeling was accomplished by a two-step synthesis, compromising a nucleophilic substitution followed by hydrolysis of the sulphamidate group. Human whole hemisphere autoradiography (ARG) was performed with [ 18F]fluororasagiline. Blocking experiments with pirlindole (MAO-A), l-deprenyl and rasagiline (MAO-B) were conducted to demonstrate the specificity of the binding. A positron emission tomography (PET) study was carried out in a cynomolgus monkey where time activity curves for whole brain and regions with high and low MAO-B activity were recorded. Radiometabolites were measured in monkey plasma using gradient HPLC. Compound 4 inhibited MAO-B with an IC 50 of 27 nM and MAO-A with an IC50 of 2.3 muM. Radiolabeling of precursor 3 and subsequent hydrolysis of the protecting group towards (1S,2S)-2-[18F]fluoro-N-(prop-2-yn-1-yl)indan-1-amine (6) was successfully accomplished with an radiochemical yield of 40-70%, a radiochemical purity higher than 99% and a specific radioactivity higher than 200 GBq/mumol. ARG demonstrated selective binding for [18F] fluororasagiline (6) to MAO-B containing brain regions, for example, striatum. The initial uptake in the monkey brain was 250% SUV at 4 min post injection. The highest amounts of radioactivity were observed in the striatum and thalamus as expected whereas in the cortex and cerebellum lower levels were observed. Metabolite studies demonstrated 30% unchanged radioligand at 90 min post injection. Our investigations demonstrated that the new ligand [ 18F]fluororasagiline (6) binds specifically to MAO-B in vitro and has a MAO-B specific binding pattern in vivo. Thus, it could serve as a novel potential candidate for human PET studies.

Methods for obtaining the trans-1-amino-2-hydroxycycloalkanes and their amides from alkenes are also disclosed, as are the novel, substantially optically pure 1-amino-2-indanols and 1-amido-2-indanols obtained thereby.
