The impact of pressure and frictional heating on retention, selectivity and efficiency in ultra-high-pressure liquid chromatography

The effects of pressure and frictional heating deserve serious consideration in ultra-high-pressure liquid chromatography (UHPLC) separations, as the pressures used can be three times greater than those in conventional high-performance LC (HPLC). We show that the effects of pressure alone can give useful selectivity effects, especially when separating molecules of different size. Frictional heating effects can cause serious losses in column efficiency and may also give changes in the selectivity of the separation. Nevertheless, the detrimental effect of frictional heating can be reduced, for instance by the judicious selection of column thermostat and particle type. In practical situations, pressure and heating effects occur simultaneously and can cause problems in transferring methods from conventional HPLC to UHPLC. In reversed-phase separations, the typical effect of increased retention with increasing pressure is counteracted by the reduction in retention that usually occurs at elevated temperatures.

Identification of Drugs Inducing Phospholipidosis by Novel in vitro Data

Drug-induced phospholipidosis (PLD) is a lysosomal storage disorder characterized by the accumulation of phospholipids within the lysosome. This adverse drug effect can occur in various tissues and is suspected to impact cellular viability. Therefore, it is important to test chemical compounds for their potential to induce PLD during the drug design process. PLD has been reported to be a side effect of many commonly used drugs, especially those with cationic amphiphilic properties. To predict drug-induced PLD in silico, we established a high-throughput cell-culture-based method to quantitatively determine the induction of PLD by chemical compounds. Using this assay, we tested 297 drug-like compounds at two different concentrations (2.5muM and 5.0muM). We were able to identify 28 previously unknown PLD-inducing agents. Furthermore, our experimental results enabled the development of a binary classification model to predict PLD-inducing agents based on their molecular properties. This random forest prediction system yields a bootstrapped validated accuracy of 86%. PLD-inducing agents overlap with those that target similar biological processes; a high degree of concordance with PLD-inducing agents was identified for cationic amphiphilic compounds, small molecules that inhibit acid sphingomyelinase, compounds that cross the blood-brain barrier, and compounds that violate Lipinski’s rule of five. Furthermore, we were able to show that PLD-inducing compounds applied in combination additively induce PLD.

Design of benzophenone-containing photoactivatable linear vasopressin antagonists: Pharmacological and photoreactive properties

We designed and synthesized new photoactivatable linear vasopressin analogues containing benzophenone photophores. All compounds were monitored and purified using RP-HPLC and characterized by mass spectrometry. Affinity and selectivity were determined in CHO cells expressing either human V1a, V1b or V2 receptor subtypes. Within the series, compounds 6 (PhCH2CO-LBpa-Phe-Gln-Asn-Arg-Pro-Arg-Tyr(3I)-NH2) and 9 (PhCH2CO-DBpa-Phe-Gln-Asn-Arg-Pro-Arg-Tyr(3I)-NH2), containing a benzoylphenylalanine residue (Bpa), were selected and their antagonistic properties determined (Kinact = 1.87 and 0.35 nM, respectively). The dissociation constant of the most potent candidate (compound 9) was further calculated from saturation experiments using the 125I derivative (Kd = 0.07 ± 0.01 nM). Photolabeling experiments using radioactive compound 9 as a probe were specific and UV-dependent and allowed the identification of two bands at molecular masses around 85-90 kDa and 46 kDa, respectively, as previously described using two photoreactive linear azidopeptide antagonists (Phalipou et al. J. Biol. Chem. 1997, 272, 26536-26544 and Phalipou et al. J. Biol. Chem. 1999, 274, 23316-23327). The results suggest therefore that compound 9 is a potent new tool for the accurate mapping of the human V1a receptor antagonist binding site.

Self-assembled fibrillar networks of a multifaceted chiral squaramide: Supramolecular multistimuli-responsive alcogels

Chiral N,N?-disubstituted squaramide 1 has been found to undergo self-assembly in a variety of alcoholic solvents at low concentrations leading to the formation of novel nanostructured supramolecular alcogels. The gels responded to thermal, mechanical, optical and chemical stimuli. Development of a novel fluorine-18 labeled deuterated fluororasagiline ([18F]fluororasagiline-D2) radioligand for PET studies of monoamino oxidase B (MAO-B)

The objective of this study was to synthesize and evaluate a novel fluorine-18 labeled deuterium substituted analogue of rasagiline (9, [ 18F]fluororasagiline-D2) as a potential PET radioligand for studies of monoamine oxidase B (MAO-B). The precursor compound (6) and reference standard (7) were synthesized in multi-step syntheses. Radiolabeling of 9 was accomplished by a two-step synthesis, compromising a nucleophilic substitution followed by hydrolysis of the sulfamidate group. The incorporation radiochemical yield from fluorine-18 fluoride was higher than 30%, the radiochemical purity was >99% and the specific radioactivity was >160 GBq/mumol at the time of administration. In vitro compound 7 inhibited the MAO-B activity with an IC50 of 173.0 ± 13.6 nM. The MAO-A activity was inhibited with an IC50 of 9.9 ± 1.1 muM. The fluorine-18 version 9 was characterized in the cynomolgus monkey brain where a high brain uptake was found (275% SUV at 4 min). There was a higher uptake in the striatum and thalamus compared to the cortex and cerebellum. A pronounced blocking effect (50% decrease) was observed in the specific brain regions after administration of l-deprenyl (0.5 mg/kg) 30 min prior to the administration of 9. Radiometabolite studies demonstrated 40% of unchanged radioligand at 90 min post injection. An efficient radiolabeling of 9 was successfully established and in the monkey brain 9 binds to MAO-B rich regions and its binding is blocked by the selective MAO-B compound l-deprenyl. The radioligand 9 is a potential candidate for human PET studies.
