Past studies showed that chemicals in cigarette smoke inhibit oviductal functioning in vivo and in vitro. The purposes of this study were to identify individual toxicants in cigarette smoke solutions that inhibit various aspects of oviductal functioning and to determine their effective doses using in vitro bioassays. Solid phase extraction and gas chromatography-mass spectrometry (GC-MS) were used to identify individual chemicals in mainstream (MS) and sidestream (SS) cigarette smoke solutions. Pyridines, which were the most abundant class of compounds identified, were purchased, assayed for purity, and tested in dose-response studies on hamster oviducts. The lowest observable adverse effect level was determined for each pyridine derivative using the oocyte pick-up rate, ciliary beat frequency, and infundibular muscle contraction assays. 2-Methylpyridine, 4-methylpyridine, 2-ethylpyridine, 3-ethylpyridine, and 4-vinylpyridine were inhibitory at picomolar concentrations in all assays. This work shows picomolar doses of pyridines with single methyl or ethyl substitutions significantly inhibit oviductal functioning raising questions regarding the safety of these compounds.

A series of simple 2-methylpyridines were synthesized in an expedited and convenient manner using a simplified bench-top continuous flow setup. The reactions proceeded with a high degree of selectivity, producing alpha-methylated pyridines in a much greener fashion than is possible using conventional batch reaction protocols. Eight 2-methylated pyridines were produced by progressing starting material through a column packed with Raney nickel using a low boiling point alcohol (1-propanol) at high temperature. Simple collection and removal of the solvent gave products in very good yields that were suitable for further use without additional work-up or purification. Overall, this continuous flow method represents a synthetically useful protocol that is superior to batch processes in terms of shorter reaction times, increased safety, avoidance of work-up procedures, and reduced waste. A brief discussion of the possible mechanism(s) of the reaction is also presented which involves heterogeneous catalysis and/or a Ladenberg rearrangement, with the proposed methyl source as C1 of the primary alcohol.

Diols exhibiting the structure (HO)CH2-(CH2) n-CH2(OH) with n = 1 up to n = 4, (HO)CH 2-CH(OH)-CH3 and cyclohexan-1,2-diols as well as cyclohexan-1,4-diol, which may establish different intramolecular interactions, are used as model substances to describe the external hydrogen bonding behaviour of multivalent hydrogen bond donors in the presence of hydrogen bond acceptor molecules. In this study, hydrogen bonds formed by the diols with tertiary aromatic amines have been investigated. In solution, different associate formation between the diols and the acceptor molecules as sketched in Fig. 1 may occur. Besides 1:1 associates formed by the interaction of one diol molecule with one amine molecule, 1:2 associates may be observed where each OH function interacts with one amine molecule. The equilibrium constants of the associates of those interactions have been studied by FTIR spectroscopy. The results allow a classification of the used diols in three different groups based on the position of the OH groups in the donor molecule. For diols with proton donating OH where no intramolecular hydrogen bond can be formed, the intermolecular hydrogen bonds for a 1:1 system may be described by thermodynamic parameters which are nearly twice the value of the corresponding equilibrium constants of monovalent alcohol systems due to the statistical weight of the OH groups. Secondly, when intramolecular hydrogen bond exists in the diols, the equilibrium constants in the interaction with the amine raise up by a factor of ca. 2-3 due to the cooperativity effect. Thirdly, if the OH groups are arranged in 1,2 positions, both OH groups may be described as independent of each other in their intermolecular interaction with the amine.
